You actually can't. There are several puzzles where you have to shoot something, and some bosses require shooting (Salazar and Saddler iirc)

I don't think it's even possible to beat El Gigante with a knife, his attacks are just too wide and Leon's too slow. If we allow grenades, then maybe you could make it to Big Cheese, but grenades don't really work on his 2nd form IIRC.
